If you have 1H Weapn Mastery, do your slays/fatalblows extend to both hands, or still just the 1 hand but for all 1H Weapons?

Nope! You can only use it in the hand that you purchased the skills in.

All profs/slays bought with 1-Handed weapons will transfer to a 2-Handed weapon, in the case of Weapon Mastery.

If you have +4 Prof/2 Slays in your primary had and +3 Prof/1 Slay in your offhand, if you pick up 2-Handed Mastery, you would have +7 Prof/3 Slays. 

It only works this way going from 1-Handed to 2-Handed.  If you go from 2-Handed to 1-Handed, all profs/slays must go to your primary hand.

Ok, another question. If I have a 1H Weapon. May I swing it with 2 hands? I know the damage would not increase and only profs from the Primary hand would apply.  I.E. I have a very long 1H sword and just for effect I want to swing it with 2 hands.

Yes, but you would not do any more damage than that weapon normally would.  For example, using a longsword with a long handle two-handed does not increase the damage to two-handed sword levels.
